Hexahydrophthalic anhydride (HHPA) is a highly sensitizing industrial chemical that is known to covalently bind to endogenous proteins.
The aim of this study was to determine the binding sites of HHPA (Hexahydrophthalic Anhydride) to human serum albumin (HSA).
Conjugates between HSA and HHPA (Hexahydrophthalic Anhydride), at two different molar ratios, were synthesized under physiological conditions.
The conjugates were digested with trypsin and Pronase E to obtain specific peptides and amino acids, which were separated by liquid chromatography (LC).
Fractions containing modified peptides were detected through quantification of hydrolysable HHPA using LC coupled to a triple quadrupole mass spectrometer with electrospray ionization.
Modified residues in albumin were identified by sequence analyses using nanoelectrospray quadrupole time-of-flight mass spectrometry.

Discussion and conclusions of HHPA (Hexahydrophthalic Anhydride):
HHPA (Hexahydrophthalic Anhydride) is primarily used as a hardener in epoxy resin systems.
HHPA (Hexahydrophthalic Anhydride) is a known respiratory sensitiser.
The critical effects of exposure include asthma, allergic rhinitis and eye and upper respiratory tract irritation.
Sensitisation is observed in workers exposed at 10 to 50 µg/m3.
Workers exposed to less than 10 µg/m3 but with intermittent peak exposures above 50 µg/m3 (five minutes per week to fifteen minutes per day) had significantly higher levels of specific immunoglobin antibodies compared to others similarly exposed but without the peak exposures.
Given that the evidence demonstrates the potential for a severe health effect resulting from acute fluctuations in airborne concentration, a peak limitation of 0.005 mg/m3 is recommended as derived by.
This concentration is cited as protective of sensitisation in exposed workers.
The present invention discloses a determination method for HHPA (Hexahydrophthalic Anhydride) content and methylhexahydrophthalic anhydride content in an electrical and electronic product.
The determination method comprises: adding a certain amount of toluene or acetone to a prepared sample; extracting a target compound in the obtained material through ultrasonic extraction or Soxhlet extraction; filtering the obtained extraction solution to obtain a sample solution; adopting a gas chromatograph-mass spectrometer (GC-MS) system to carry out separation detection; and adopting an external standard method to carry out quantitation.
According to the present invention, with the systematic comparison study, the gas chromatography-mass spectrometry detection method for the HHPA (Hexahydrophthalic Anhydride) content and the methylhexahydrophthalic anhydride content in the electrical and electronic product is established; and the method has technical characteristics of rapidness, efficiency, good separation effect, accurate determination, strong specificity, simple and rapid analysis, and the like.

Post-shift and next-morning urine was sampled from workers exposed to hexahydrophtalic anhydride (HHPA), an epoxy hardener, sensitising at low exposure levels.
Exposure levels of HHPA (Hexahydrophthalic Anhydride) in air gas chromatography, GC) in the range of 30–270 μg/m3 corresponded to urinary concentrations of 0.9–2.8 μmol hexahydrophthalic acid (HHP acid; GC-mass spectrometry)/mmol creatinine.
In the morning samples the concentrations were <0.04-0.3 μmol HHP acid/mmol creatinine.
In unexposed controls, the level was <0.1 μmol/mmol creatinine.
A correlation was found between the time-weighted levels of HHPA (Hexahydrophthalic Anhydride) in air and HHP acid in the post-shift urine (r s = 0.93; P < 0.023), indicating that the determination of HHP acid in urine is suitable for biologic monitoring of HHPA exposure.
